In living macaques, CRISPR was used to edit ~66% of liver cells by introducing a single nucleotide mutation in the gene PCSK9. This caused their LDL cholesterol to drop by 60% and stay that way for at least 8 months (no further tests yet).
Off-target mutations were only at one site with a frequency of <1%. In humans, a similar mutation in PCSK9 would lead to a reduced chance of developing atherosclerosis-related diseases.
The entire procedure consisted of a single injection of lipid nanoparticles containing CRISPR into the blood.
Here was used a variant of CRISPR which can replace only one base, without double-stranded DNA breaks.
